Genetic differentiation between wild and cultived Momordica charantia: evidence of SSR and chloroplast DNA variation.

Using the genetic variation of SSR fingerprinting and the sequence variations of two chloroplast non-coding regions, including the atpB to rbcL and the trnL to trnF spacers, were estimated the genetic variation among the different varieties of Momordica charantia. In this study, we used seven wild populations, twenty cultivated varieties and two hybrid strains to sequence and analysis the genetic diversity and phylogenetic pattern. Sequencing length was detected from 953 to 954 bps in the atpB to rbcL spacer and 500 to 506 bps in the trnL to trnF spacer. Nucleotide diversity was detected 0.00210 in the atpB to rbcL spacer and 0.00552 in the trnL to trnF spacers, showing higher levels of genetic variation in the trnL to trnF spacer than atpB to rbcL spacer. Combined the atpB to rbcL and the trnL to trnF spacers, we got 1454-1459 bps in the sequencing length and 0.00261 in the nucleotide diversity. A reconstructed NJ tree and nest clade tree by using combined data showed the three major groups, including one group of wild populations and two groups of cultivated varieties. Two groups of cultivated varieties related to the morphological character of fruit color, one was green and another was white. Two hybrid strains in the gene genealogy demonstrated to the group of wild populations showed the maternal inheritance from wild parental individuals. Based on 11 polymorphic SSR loci, this is highly consistent with the AMOVA analysis which showed that 95% of the variation existed within groups and 5% of the variance existed among groups. *All authors contributed equally to this work
