SnRK1 activity during phosphate deficiency in Arabidopsis thaliana.

Phosphate (Pi) is one of the essential nutrients required by plants for growth and development. During Pi deficiency multiple changes occur at the morphological and biochemical levels to enhance Pi availability. There is a considerable scientific interest in understanding how plants sense and response to Pi deficiency. Several genes encoding proteins involve in signal transduction have been identified during Pi deficiency. In bean plants, one of these genes encode a protein with high homology to the catalytic subunit of the animals AMPK. Animals AMPK, yeast SNF1 and plants SnRK1 belongs to the AMPK/SNF1/SnRK1 family of protein kinases which are involved in controling metabolic pathways in response to energy status. The active kinase is formed by three different subunits: a catalytic alpha subunit and two regulatory beta and gamma subunits. In Arabidopsis thaliana, there are three different genes encoding catalytic subunits (AKIN 10, AKIN 11 and Mul 8) and has been hypothesized that, as in animal systems, each subunit might be part of a heterotrimeric complex with a particular function. We evaluated SnRK1 activity in Arabidopsis plants grown during 5 and 10 days on Pi-deficient conditions. Results indicated that since 5 days of Pi deficiency there was a decrease in SnRK1 activity, although gene expression remains constant during the same period of time. In order to know which of the catalytic subunit was down regulated by Pi deficiency, we developed Arabidopsis transgenic plants that overexpressed AKIN10 and AKIN 11 GFP fusion proteins. Analysis and characterization of these plants during Pi deficiency will be presented.
