Regulation of mitogen activated protein kinases (MAPKs) and MAPK kinase (MAPKKs) in rice in response to various abiotic stresses.

Mitogen activated protein kinases (MAPK) cascades are known to play a crucial role in stress tolerance and are evolutionary conserved signaling modules in yeast, animals and plants. MAPK cascades are composed of three protein kinase modules MAPKK kinases (MAPKKKs), MAPK Kinases (MAPKKs) and MAPKs which are linked in various ways to up stream receptors and down steam targets. In order to understand the function of different MAPKs and MAPKKs in rice we attempted to clone the entire members of the gene family using reverse- transcriptase polymerase chain reaction (RT-PCR) in indica-cultivar group, var: Pusa Basmati-1. We here report transcript regulation of different members of MAPKs and MAPKKs in response to various abiotic stresses. Quantitative real time polymerase chain reaction (qRT-PCR) revealed differential regulation of different members of theses protein kinase family. The activation of MAPK activity as analyzed by in-gel assay, revealed fast activation of the protein activity in response to heat, cold, salinity and drought stress. Interestingly keeping the plants in stress for longer duration resulted in the steady state elevated MAPK activity for a period of 24 hours with certain rhythm in case of cold and heat stress. While moving back the plant to normal condition after giving a shock of high or low temperature resulted in fast and transient activation of MAPKs. Yeast two hybrid interaction studies revealed that MAPKKs interacts with specific MAPKs and that this interaction is stress specific. In order to identify the specific members of these protein kinase families being activated by different stresses, antibodies directed against poorly conserved regions of a few of MAPKKs and MAPKs is being generated. The results obtained will be discussed.
