We are interested in evaluating the potential health risks and/or benefits of phytoestrogens in soy-foods. Here we will report on our progress in breeding for isoflavone-free (phytoestrogen-free) soybean. This project shows how TILLINGĀ® (Targeted Induced Local Lesions In Genomes) assisted breeding can be used to modify a complex trait by creating an allelic series of mutations in targeted genes. The synthesis of isoflavones involves at least two isoflavone synthase genes (IFS1 and IFS2). We have used TILLINGĀ® to select plants carrying IFS1 and IFS2 nonsense and missense mutations resulting in no, or reduced, IFS activity. We will present genotype and phenotype data for plants carrying mutations in either or both of these genes and evaluate the contribution of each IFS gene to the level of isoflavones.
