Regulation of organ boundaries during reproductive development in Arabidopsis.

Post-embryonic development in flowering plants is regulated by the shoot apical meristem (SAM), which maintains a population of pluripotent stem cells. A key stage during plant development is the transition from vegetative to reproductive phase. During the floral transition in Arabidopsis, the vegetative SAM changes into the inflorescence meristem (IM). Upon the transition, the SAM switches from making rosette leaves to initiating lateral organs. Therefore, it is essential to separate the distinct patterns of gene expression that maintain stem cells from those that regulate differentiation of lateral organs. This is accomplished by lateral organ boundaries that are established and maintained between the SAM and the newly forming primordia. However, the mechanisms that regulate such boundaries are poorly understood. Studies haves shown that two redundantly functioning genes PENNYWISE (PNY) and POUNDFOOLISH (PNF) are important in internode and floral cell fate specification. Moreover, in pnyPNF/pnf mutants pedicels are often fused to each other and to the main stem, demonstrating the role of PNY/PNF in maintaining boundaries between IMs and floral meristems (FMs). Our goal was to study how PNY/PNF might function with other known boundary genes, such as, LATERAL SUPPRESSOR (LAS) and CUP-SHAPED COTYLEDON (CUC). Preliminary results indicate that loss of LAS function in pnyPNF/pnf mutants enhance the boundary defects. These results indicate that LAS and PNY/PNF might be functioning independently to regulate organ separation during reproductive development. We are currently analyzing possible interactions between PNY/PNF and LAS using yeast two hybrid. Further results from this study will be presented at the society meeting.
