Two structural variants of the CONSTANS-LIKE gene in long- and short-day Solanum plants.

CONSTANS and its CONSTANS-LIKE (COL) orthologs have been established as the key gene in the photoperiodic control of floral transition in arabidopsis, rice and several other plant species. Comparative analysis of COL clones from long-day (LD) Solanum tuberosum ssp. tuberosum (EF143437-EF143451, DQ499754, DQ472734) and short-day (SD) S. demissum (EF143445, DQ499755), S. stoloniferum (EF143433-EF143436) and S. tuberosum ssp. andigena (EF143437-EF143440) discerned two variants, which differed in the structure of exon 2, particularly, in the numbers of AAC/AAT and CAA/CAG repeats corresponding to polyasparagine and polyglutamine motifs in the middle region of the CONSTANS protein. The length of these repeats may affect the conformation of the CONSTANS protein and in this way modify the stability and activity of this transcription factor. Real-time PCR analysis demonstrated that both COL variants were present in 1:1 proportion in the genomes of each of four Solanum species independent of their photoperiodic reaction. However, the diurnal expression rhythm of two COL variants dramatically differed in each species, and the expression patterns of each COL variant were dissimilar in LD and SD plants.
