Phylogenetic Studies of Disterigma: a Total Evidence Approach Exploring Transformation Costs with Direct Optimization and Multiple Alignments.

The phylogenetic position and monophyly of the neotropical blueberry genus Disterigma (Ericaceae) is examined in the light of morphological and molecular evidence (ITS and ndhF data). Representatives from each of the major clades of the tribe Vaccinieae, where Disterigma is placed, are studied using a total evidence approach and maximum parsimony. Employing direct optimization as implemented in POY4, the costs of morphological change, transitions, transversions, gap opening, and gap extension are explored in a combination of twenty-nine different costs scenarios. Additionally, the results obtained with direct optimization are compared to those obtained using the multiple alignment produced by Clustal with the same costs regime.
A monophyletic Disterigma s.s. clade including more than two thirds of the species was recovered. Disterigma s.s. is characterized by a pair of bracteoles well differentiated from other bracts in size and position, which envelope the calyx; its flowers are 4(-5) merous; and its stamens are all equal in size and twice the number of floral parts. The segregation of some species from Central America and the central Andes from Disterigma s.l. is supported by all the analyses and individual data sets, however, their true affinities remain unknown.
