Chemical Communications: Deciphering the chemical language of C. elegans.

Living organisms release a number of chemical compounds to their environment, some of which are used in chemical communication. The goal of this study is to identify molecules produced by Caenorhabditis elegans, a small soil nematode and a model organism, in order to begin to decipher its chemical language. Conditioned water of life stages (Egg, L1, L2, L2D, L3, L4, Young Adult and Adult) of C. elegans was analyzed to see whether each life stage produces a signature of compounds. A 1-mm triple resonance high temperature superconducting (HTS) probe at 600 MHz was used for nuclear magnetic resonance (NMR) analysis of worm conditioned water. The probe has a sensitivity that is about 25x greater than a conventional 5-mm probe at the same field strength, and a minimum total sample volume of just 8 µL (Brey et. al., 2006). Based on two dimensional NMR, there are common resonances to all of the life stages and unique resonances to each life stage. Compounds that are released to the water will be separated by various chromatographic methods, tested for biological activity, further purified, and identified by mass spectrometry and NMR. This information will not only add to the extensive scientific knowledge of C. elegans but may provide important new clues for the biological control of other nematode parasites of plants or animals.
