Phylogeny of Isoetes ×bruntonii as revealed by spore morphology, orbicule typology, intercellular pectic protuberances and DNA sequences.

Based on its morphology and distribution, Isoetes ×bruntonii Knepper & Musselman, (2n = 3x = 33) has been suspected to be an allotriploid derivative of the basic diploid (2n = 2x = 22) I. engelmannii A. Br. and the allotetraploid (2n = 4x = 44) I. hyemalis D. F. Brunton. In the inner coastal plain and adjacent piedmont regions of Virginia, the allotriploid occurs with both putative parents in shallow depressions and adjacent slopes of both intermittent and permanent blackwater streams. Examination of this species complex provides an opportunity to describe the effect of interspecific hybridization on morphological and ultra-structural characters (and their character states) considered to be useful in distinguishing the taxa involved. In this regard, we describe spore morphology, orbicule typology, and the form and texture of intercellular pectic protuberances using scanning electron microscope images. Preliminary DNA analysis of the second intron of a LFY homolog is being generated to help corroborate the morphological, cytogenetic and anatomical data.
