Hormone interaction and control responses of different Agave explants (meristem, buds and leaf).

Maguey or Agave pulquero (Agave atrovirens and A.  salmiana), is the important crop in developing countries. These are the largest agave plants in the world. The agave sap, known as aguamiel is obtained by scratching the medulla in the agave stem. The sap is then fermented for the production of alcoholic and slightly acidic traditional beverage. The improvement of agave pulquero is vital as the productivity is adversely affected by abiotic stresses and fungal infections. The multiplication rate of this plant is very low, the sexual propagation by seed, involves a period of 12 to 15 years, which makes unfeasible any program for accelerated reproduction. In contrast, the work on propagation of this plant using the techniques of plant tissue culture has been reported for s some Agave species. The development of a reliable plant regeneration protocol for this crop plant will for the basis for its genetic improvement program. In this study we established the response of the meristem, bud and leaf base of agave plants as sources for explants in medium MS with hormones (2,4-D, Kinetin, 6-BAP and TDZ). Explants were tested for the production of callus and shoot-forming.
CALLUS-FORMING: 2,4-D and 6-BAB at the equal concentration was the best mix in callus formation when leaf base was used as explant, (83% callus formation). We analyze different mix for develop callus tissue. The mix where de callus was big and vigorous was 0.25mg/l 2,4-D and 1.0mg/l 6-BAP. SHOOT-FORMING. In lateral buds explants, the adventitious shoots were directly developed from the protuberances. The induction rate of shoot was influenced by unique hormone: Kinetin (1mg/l) or 6-BAP (1mg7l) or TDZ (1-3mg/l). When we use apical meristem explants, we recognized three different parts: high, medium and low. The medium part was the best explant for promoting shoot and little plantlet.
