Oxidative Stress

A tomato zinc finger protein associate with environmental stresses and affect the developmental stage in Arabidopsis.

Many of plant genes can response to various stresses and might function to fit to the change of environment. Previously, we have identified several genes that would response to various stresses in our home-made tomato cDNA microarray. One of these genes, cLEX4I22, which would response among water deficit, salt and chilling, was selected for the functional study. We firstly identified the full length cDNA by Rapid amplified cDNA ends (RACE) and blast result shown it belongs to C2H2 type zinc finger proteins. Genomic PCR to amply full length CDS and DNA sequence indicated it might contain no intron. Onion bombardment of 4I22/GUS fusion protein has shown that they can localize to nucleus. Over-expression 4I22 in Arabidopsis display dwarf, shorter root length and silique while it seems normal as using Rd29a inducible promoter to drive 4I22. However, the 4I22 proteins are expressed by Rd29a promoter, the root length phenomena will be observed in these transgenic plants treated with osmotic stress. To understand the relationship of 4I22 to environmental stresses, WT and 4I22 over-expression Arabidopsis lines were compared. Interestingly, over-expression 4I22 seems to be more sensitive to freezing, water deficit, and salt condition. These results indicated that 4I22 in Arabidopsis might be influence on development and also associate with environmental stresses.
