Growth and Vegetative Development

Development of a linkage genetic map for chilling requirement in peach.

Chilling requirement to release endodormancy in the vegetative and floral buds of temperate trees is a quantitative trait of ecological and economic importance. We are developing a genetic linkage map for chilling requirement in peach (Prunus persica L. [Batsch]) using two populations derived from independent high and low chill requiring parents. Two F2 progenies were created for mapping and phenotyping. The first cross was between ‘Contender’ peach (high chill) × ‘Fla92-2C’ (a low chill selection). This F2 population consists of 378 trees and also segregates for the qualitative non-showy (Sh)/showy (sh) petal-size phenotype. The second cross is ‘Hakuho’ (high chill) × ‘UF Gold’ (low chill) and this F2 population consists of 57 trees. Both populations were screened for polymorphisms using 259 SSR (simple sequence repeat) markers. Following screening, 51 markers with distribution across all eight P. persica linkage groups were used to construct an SSR framework map based on the general Prunus linkage group map. Flower bud flush date of F2 individuals was scored in the spring of 2006 and 2007 and segregates in a continuous fashion. F2 individuals of the ‘Contender’ × ‘Fla92-2C’ cross were scored as non-showy (Sh) or showy (sh) in the spring of 2006 and displayed the expected 3:1 ratio of Sh:sh phenotype. The Sh/sh trait mapped to LG8. This is the first time that the Sh/sh flower trait has been genetically mapped and it is the first phenotypic trait to be mapped to LG8 in Prunus. We are currently saturating the map with AFLP markers and conducting forcing experiments to evaluate the quantitative chilling requirement of each F2 individual.
