Scarecrow (SCR) Mediates Root Development in Lupinus albus and Medicago truncatula. EST sequencing of white lupine (Lupinus albus) phosphorus stressed cluster roots revealed the expression of a scarecrow like gene (LaSCR), belonging to the Gras family of transcription factors. Transcript abundance as measured by RT-PCR and RNA blots was low in all tissues assayed. SCR is known to regulate the asymmetric cell division required for the formation of ground tissues (cortex/endodermis) during root development in many species. In order to determine the biological function of SCR in L.albus and Medicago truncatula we developed a RNAi construct for delivery to roots via Agrobacterium rhizogenes transformation. The RNAi construct was made using a 450 bp segment of the LaSCR coding region. Medicago truncatula root development was impaired upon suppression of LaSCR expression.
