Plant-Symbiont Interactions Jauregui, David [1], Nava, Noreide [2], Alvarado, X [2], Santana, O [2], Quinto, Carmen [2]. A leucine rich repeat receptor-like kinase has a role in nodule morphogenesis in Phaseolus vulgaris. Establishment of symbiosis between legume plants and nitrogen-fixing bacteria is a complex process, orchestrated by an exchange of signal molecules, developmental and gene expression changes from both organisms. During the nodulation process several genes that are essential for the early stages of this symbiotic interaction, such as those involved in signal perception and transduction have been identified in legumes. Among these genes, receptor-like kinases (RLKs) have been found (1, 2), which encode for a unique motif with three leucine-rich repeats (LRR) in the extracellular domain, a transmembrane helix and an intracellular serine-threonine kinase domain. Previously, in our laboratory a cDNA encoding a LRR-RLK was isolated from a cDNA library of P. vulgaris roots (PvRLK). The extracellular domain of PvRLK was cloned and expressed in Escherichia coli as a His-tagged protein. Herein, the immunolocalization of PvRLK with anti-PvRLK polyclonal antibodies is showed. Data will be presented suggesting that this protein is involved in the vascularization of the nodule. Additionally, a 3D model of the extracellular domain of the presumed receptor including the LRR domain was done in silico to establish a possible structure-function relationship. Furthermore, biochemical studies are in progress to identify putative ligands and PvRLK associated proteins that may be participating in the signaling process. 1. Endre, et al. 2002.
