Examining uncertain phylogenetic relationships within section Nycterinia of genus Zaluzianskya (Scrophulariaceae s.s., tribe Limoselleae) using DNA sequences from the chloroplast genome. The southern African flowering genus Zaluzianskya (Scrophulariaceae s.s.) consists of approximately 55 species which have been grouped into four sections: Zaluzianskya, Macrocalyx, Holomeria, and Nycterinia. In previous phylogenetic hypotheses, there has been some uncertainty in the placement of Z. mirabilis and Z. microsiphon. Previous analyses of trnL-F and rpl16 of the chloroplast genome placed Z. mirabilis of section Macrocalyx within a well-supported clade of species from sect. Nycterinia. In contrast, analyses of nrDNA ITS placed Z. mirabilis outside of this clade, leaving a monophyletic Nycterinia clade. Additionally, several populations of Z. microsiphon, the sole day-flowering species within section Nycterinia, did not form a clade in previous estimates of phylogeny. The present study used sequences from the cpDNA gene regions rps16 and trnS-fM to infer phylogeny for Zaluzianskya section Nycterinia. Relationships inferred by these two cpDNA regions were consistent with those suggested by trnL-F and rpl16. Zaluzianskya mirabilis is placed (with strong support) within the Nycterinia clade, despite its contrasting morphology, habitat, and distribution. This study greatly increased the sampling of Z. microsiphon (with 21 populations) and found strong support for the division of this species into at least two separate lineages placed in different regions of the Nycterinia clade.
