A Chemical Genetics Approach for Dissection of Abscisic Acid Signaling Network.

Using GFP reporter transgenic lines under the control of a strong ABA-inducible promoter, we have screened a chemical library of 9,600 compounds to identify small molecules inhibiting ABA-induced gene expression. One particular small molecule was found to specifically block ABA-dependent gene expression. In addition, this chemical compound inhibits ABA- and Ca2+-induced stomatal closure while minimally affecting an ABA-delayed seed germination phenotype. Transcriptome analyses show that this compound strongly inhibits ABA induction of more than 25% of the ABA-inducible genes, some of which were confirmed by individual quantitative PCR analyses. To dissect further ABA signal transduction mechanisms and to understand the physiological role of the chemical compound, forward genetic screens have been pursued for mutants resistant to the compound’s inhibitory effect on ABA-induced gene expression. Examples of isolated mutants with strong ABA signaling and guard cell phenotypes will be presented and advantages of chemical genetics as a tool to study the robust ABA signaling network will be discussed.
